In order to fully benefit from the resolving power of single molecule localization techniques, it is very important to minimize aberrations that arise in real life situations.<\/p>\n

The microscope\u2019s components, the refractive index mismatch between the objective and the immersion medium, and the biological sample itself can all introduce various types of aberrations.<\/p>\n

This results in a degradation of the PSF\u2019s quality which leads to a reduction in the number of detected photons and, ultimately, a reduction in resolution.<\/p>\n
